Potensi Limbah Kulit Salak Sebagai Pewarna Hematoxylin Alami Preparat Cairan Pleura

Pewarnaan sitologi cairan pleura merupakan langkah penting dalam diagnosis berbagai penyakit, terutama keganasan, dengan Hematoxylin sebagai pewarna inti yang umum digunakan. Namun, Hematoxylin memiliki keterbatasan berupa potensi toksisitas, harga relatif tinggi, serta dampak lingkungan. Penelitian ini bertujuan untuk mengevaluasi pemanfaatan limbah ekstrak kulit salak (Salacca zalacca) sebagai alternatif pewarna Hematoxylin pada preparat cairan pleura. Penelitian dilakukan menggunakan desain eksperimen semu dengan rancangan pretest–posttest control group, membandingkan pewarnaan Papanicolaou menggunakan Hematoxylin standar dan ekstrak kulit salak pada berbagai konsentrasi (100%, 80%, 50%, dan 25%). Parameter yang dinilai meliputi intensitas warna, kontras, kejernihan, keseragaman pewarnaan dan kestabilan pewarnaan yang dievaluasi secara blind oleh dua analis dan menggunakan uji Chi-Square. Hasil menunjukkan bahwa ekstrak kulit salak konsentrasi 100% memberikan kualitas pewarnaan terbaik dan tidak berbeda signifikan dengan Hematoxylin standar (p = 0,479), sedangkan konsentrasi lebih rendah menunjukkan penurunan kualitas pewarnaan. Kesimpulannya, ekstrak limbah kulit salak berpotensi digunakan sebagai alternatif pewarna Hematoxylin yang lebih aman, murah, dan ramah lingkungan pada preparat cairan pleura.
