Abstrak

Exploring the Antioxidant and Anti-Inflammatory Properties of Forest Honey from Buton Island through In Vitro Assays and In Vivo Edema Inhibition Tests

Forest honey is a natural product derived from the nectar of wild plant species and contains various bioactive constituents, including flavonoids and phenolic compounds, that contribute to its antioxidant and anti-inflammatory activities. Buton Regency in Southeast Sulawesi is known as a producer of high-quality forest honey; however, scientific evidence regarding its biological properties remains scarce. This study aimed to investigate the free radical scavenging capacity and anti-inflammatory effects of forest honey from Buton using both in vitro and in vivo approaches. Antioxidant activity was evaluated through DPPH and ABTS assays, while anti-inflammatory effects were assessed via BSA protein denaturation inhibition and a xylene-induced mouse ear edema model. The honey demonstrated IC₅₀ values of 71.69 ± 2.73 mg/L (DPPH) and 49.23 ± 4.24 mg/L (ABTS), and an IC₅₀ of 14.17 ± 3.18 mg/L for protein denaturation inhibition. In vivo, high-dose administration significantly reduced ear edema, with inhibition levels comparable to diclofenac sodium. Statistical analyses confirmed significant differences across most parameters (p < 0.05). These results indicate that Buton forest honey possesses promising antioxidant and anti-inflammatory properties, supporting its potential development as a natural therapeutic agent.

Madu hutan merupakan produk alami yang dihasilkan dari nektar tumbuhan liar dan kaya akan senyawa bioaktif seperti flavonoid dan fenolik, yang diketahui memiliki potensi sebagai antioksidan dan antiinflamasi. Kabupaten Buton, Sulawesi Tenggara, dikenal sebagai salah satu daerah penghasil madu hutan, namun kajian ilmiah terkait aktivitas biologisnya masih terbatas. Penelitian ini bertujuan untuk mengkaji aktivitas antioksidan dan efek antiinflamasi madu hutan Buton secara in vitro dan in vivo. Uji antioksidan dilakukan menggunakan metode DPPH dan ABTS, sedangkan aktivitas antiinflamasi diuji melalui metode inhibisi denaturasi protein BSA serta model edema telinga mencit yang diinduksi oleh xylene. Hasil penelitian menunjukkan bahwa madu hutan Buton memiliki nilai IC₅₀ sebesar 71,69 ± 2,73 mg/L pada uji DPPH dan 49,23 ± 4,24 mg/L pada uji ABTS. Pada uji denaturasi protein, nilai IC₅₀ yang diperoleh sebesar 14,17 ± 3,18 mg/L. Secara in vivo, pemberian madu dosis tinggi mampu menurunkan ketebalan telinga mencit secara signifikan, dengan persentase inhibisi yang mendekati efek dari natrium diklofenak. Uji statistik menunjukkan bahwa sebagian besar parameter menunjukkan perbedaan yang signifikan (p < 0,05). Dengan demikian, madu hutan Buton berpotensi dikembangkan sebagai agen antioksidan dan antiinflamasi alami.
